Assessment of the MRI and Behavioral Test Results in a Focal Cerebral Ischemia-Reperfusion Model in the Rat after Separate and Combined Use of Mouse-Derived Neural Progenitor Cells, Human-Derived Neural Progenitor Cells and Atorvastatin.

OBJECTIVE
To assess the efficacy of neural progenitor cells (NPC) transplantation on ischemic stroke, and to investigate whether atorvastatin improve the therapeutic potential of NPC after stroke.
METHOD
Model of focal cerebral ischemia-reperfusion performed by temporary occlusion of the middle cerebral artery. Mice were assigned at random to receive a transplant of intracerebral mouse NPC alone (mNPC), NPC man alone (hNPC), mouse NPC plus atorvastatin oral (mNPC + A), human NPC plus atorvastatin oral (hNPC + A), atorvastatin oral, or intracerebral Dulbecco ‘s Modified Eagle’ s media injection (control group). adhesive removal, rotarod, test cylinder, and magnetic resonance imaging (MRI) is used for the assessment of rats for 4 weeks. RESULTS
The hNPC and mNPC groups showed significantly improves functional outcomes and reduced infarct area ratio compared with the control group. HNPC group had a significantly better performance and the ratio of infarct area lower than mNPC group. In addition to atorvastatin for stem cell therapy significantly improved functional outcome, even though it did not affect the ratio of infarct area on MRI. anti-inflammatory response in the area of ​​infarction was higher in the group mNPC. NPC transplantation significantly reduced the number of microglia and a significant increase in the number of astrocytes. and granzyme B lymphocyte activity CD8a + T was not detected in any of the subjects.
CONCLUSION
HNPC and mNPC both treatments significantly improved functional outcomes, and reduce the ratio of infarct area after stroke. Atorvastatin improve the therapeutic potential of NPC, including neurological improvement.

Evaluating the Wistar rats and BALB / c mice as an animal model for congenital toxoplasmosis brain and eyes.

This study was conducted to evaluate the production potential of the cyst by Toxoplasma (T.) gondii, RH strain, in Wistar rats and BALB / c mouse and the goal of this study was to introduce a suitable animal model for congenital, brain, and eyes toxoplasmosis. Rats and mice, considered a model of toxoplasmosis of the brain and eyes, the intraperitoneal infected by a different number of parasites and their eyes and brain were evaluated for the presence of T. gondii cysts using microscopic examination and bioassay methods.

Additionally, pregnant rats and mice, considered a model of congenital toxoplasmosis, which intraperitoneal infected by a different number of parasites and their babies checked by the method mentioned above. The best results for the model of cerebral toxoplasmosis was observed in mice infected with 107 parasites, so that all infants (100%) were infected with parasites when examined using bioassay method.

Furthermore, the best results were observed for brain congenital toxoplasmosis models with 100% infection rate in infants born to mothers infected with 107 parasites. Overall, only a few samples positive ocular using bioassay method. The best results in this study is to congenital toxoplasmosis brain model where pregnant mice infected with 107 parasites and all infants are infected (100%). Therefore, this baby can be used as a model of congenital toxoplasmosis brain when they are in the fetal stage, and can be used as a model of brain toxoplasmosis one month after birth.

BACKGROUND
Atsttrin, protein engineering consists of three tumor necrosis factor receptor (TNFR) -binding fragments of progranulin (PGRN), demonstrated therapeutic effect in a murine model of inflammatory arthritis. In addition, intra-articular delivery of PGRN protect against osteoarthritis (OA) developments. The purpose of this study was to determine whether Atsttrin also have a therapeutic effect on OA and molecular mechanisms involved.

METHOD
Surgically induced OA and broke noninvasive models established in the mouse and rat, respectively. OA cartilage degradation and evaluated using safranin O staining, immunohistochemistry, and ELISA. Moreover, the expression of markers associated pain, degenerative factors, and the anabolic and catabolic markers known to be involved in OA analyzed. Furthermore, anabolic and anti-catabolic effects and the underlying mechanism of Atsttrin determined using in-vitro tests with primary chondrocytes.

RESULTS
Here, we find Atsttrin effectively prevent accelerated OA phenotypes associated with PGRN deficiency. Additionally, Atsttrin shows preventive effect on OA by protecting articular cartilage and reduces pain associated OA in both mice induced nonsurgically and surgically induced murine model of OA. Mechanistic studies revealed that Atsttrin TNFR2-Akt-stimulated Erk1 / 2-dependent chondrocyte anabolism, while inhibiting TNF / TNFR1-mediated catabolism inflammation.s

Description: The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Human IDE. Standards or samples are added to the appropriate microtiter plate wells then with a biotin-conjugated antibody specific to Human IDE. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Human IDE, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Human IDE in the samples is then determined by comparing the OD of the samples to the standard curve.

CONCLUSION
These findings not only provide new insights into the role of PGRN and engineered proteins derived Atsttrin in homeostasis and OA cartilage in vivo, but also may lead to new therapeutic alternatives for OA and other degenerative joint disease relative.
